Phleomycin is a glycopeptide antibiotic structurally similar to bleomycin isolated from Streptomyces rotatum. The mechanism of action is to destroy the integrity of the double helix by binding and inserting DNA, effectively acting on most bacteria, filamentous fungi, yeast, plant and animal cells.
Putrethromycin was used as a selective antibiotic in molecular genetics experiments. Resistance to sahuthromycin is given by the Sh ble gene from the hindustani strain, which encodes a protein that binds to sahuthromycin to destroy its dna-cutting activity. Sapromycin is recommended for cells that are less sensitive to Zeocin, such as filamentous fungi and certain yeasts.
Phleomycin is provided in a sterile liquid at a concentration of 20mg/ml. The recommended concentration is 10 g/ml for yeast and 25-150 g/ml for filamentous fungi.
