Endopeptidase or endoproteinase are proteolytic peptidases that break peptide bonds of nonterminal amino acids (i.e. within the molecule), in contrast to exopeptidases, which break peptide bonds from their end-pieces. For this reason, endopeptidases cannot break down peptides into monomers, while exopeptidases can break down proteins into monomers.
Endopeptidase K is a powerful proteolytic enzyme isolated from candida albicans, which has high specific activity and is a key reagent for DNA extraction. The enzyme is active in a wide pH range (4 ~ 12.5) and at high temperature (50 ~ 70°C) for the separation of plasmid or genomic DNA and RNA. In DNA extraction, the main role is the enzymatic hydrolysis of histones bound to nucleic acid, making the DNA free in the solution, and then using different methods to extract, remove impurities and collect DNA. Neither chelating agents such as EDTA nor detergents such as SDS can deactivate them. Endopeptidase K storage solution is usually 10mg/ml or 20mg/ml. Store at a temperature of 20°C. Endopeptidase K solution is colorless and transparent, and cannot be used if precipitation occurs.
Endopeptidase K in situ hybridization technique is usually used in hybrid before processing, it have the function of the target DNA protein digestion surrounded, to increase the target nucleic acid probe and combined with the opportunity, improve the hybridization signal. But the concentration of proteinase K, digestion time is too long, or high incubation temperature too high, will have certain damage, the structure of cells in tissue section of loss, the loss of the nucleus, which affect hybrid results. EDTA buffer can replace the role of proteinase K, solve the above problems, and can achieve ideal dyeing effect.
