Description:
Dexamethasone is a potent synthetic member of the glucocorticoid class of steroid drugs. It acts as an anti-inflammatory and immunosuppressant. It is 20 to 30 times more potent than the naturally occurring hormone hydrocortisone and 4 to 5 times more potent than prednisone.
Dexamethasone was white or white crystalline powder. Odourless. Dexamethasone is slightly dissolved in methanol, ethanol, acetone or dioxane, slightly dissolved in trichloromethane, very slightly dissolved in ether, and almost insoluble in water.
Dexamethasone was used for precise weighing, dissolved in methanol and quantitatively diluted to make a solution containing 0.5mg per 1ml as the sample solution. Another betamethasone reference product was used for precise weighing, dissolved in methanol and quantitatively diluted to make a solution containing about 0.5mg per 1ml. The precise quantity was 1ml, placed in a 100ml measuring bottle, and the test product solution was added 1ml, diluted to scale with methanol, shaken well, and used as the control solution. According to the chromatographic conditions of the content determination, 20 mul of the control solution was injected into the liquid chromatograph, and the detection sensitivity was adjusted to make the peak height of dexamethasone chromatographic peak about 20% of the full range. The sample solution and the control solution were then precisely measured and injected into the liquid chromatograph, and the chromatographic chart was recorded to 2.5 times the retention time of principal component peak. If the chromatographic peak corresponding to betamethasone in the control solution appears in the chromatographic diagram of the sample solution, the peak area shall be calculated according to the external standard method, and the content shall not exceed 0.5%. The peak area of other impurities should not be greater than that of dexamethasone in the control solution (1.0%), and the sum of the peak area of all impurities should not be greater than 2 times that of dexamethasone in the control solution (2.0%). Any peaks smaller than 0.01 times of dexamethasone peak area in the control solution in the chromatographic diagram of the sample can be ignored.
