Description:Amikacin is an aminoglycoside antibiotic, consisting of a white or a similar white powder or a crystalline powder; Almost odorless, tasteless. Its mechanism of action is to act on ribosomes in bacteria, inhibit the synthesis of bacterial proteins, and destroy the integrity of bacterial cell wall, leading to the destruction of bacterial cell membrane and cell death. It is suitable for infection caused by kelan-negative bacillus and staphylococcus aureus resistant to penicillin. For intramuscular injection or intravenous infusion, the half-life of intravenous infusion is about 2h, and it can be as long as 30h in the absence of urine, rarely binding with protein. The main cause of cochlear nerve damage, ear, renal toxicity and gentamicin similar.
Take an appropriate amount of Amikacin, add water to dissolve and dilute quantitatively to make a solution of about 10mg per 1ml; In addition, an appropriate amount of amicabine reference product was taken, accurately weighed, dissolved in water and quantitatively diluted to make a solution containing about 100 mug per 1ml. The above two solutions and water were precisely measured at 0.2ml each, respectively treated by the derivatization method of the system applicability test solution under the content determination item, and used as the sample solution, control solution and blank solution. According to the chromatographic conditions under the content determination, 20 mul of the control solution was injected into the liquid chromatograph, and the detection sensitivity was adjusted so that the peak height of the principal component chromatograph peak was about 50% of the full range, and then precise measurement was taken and 20 mul of each of the above three solutions, respectively injected into the liquid chromatograph, and the chromatographic diagram was recorded to 4 times the retention time of the principal component peak. If there is an impurity peak in the chromatographic diagram of the sample solution, the peak area of impurity A shall not be greater than the main peak area of the control solution (1.0%), the peak area of other single impurities shall not be greater than the main peak area of the control solution (1.0%), and the sum of the peak area of other impurities shall not be greater than 3 times the main peak area of the control solution (3.0%). Any chromatographic peak smaller than 0.1 times of the main peak area of the control solution, the chromatographic peak in the blank solution and the peak before the main peak in the chromatographic diagram of the sample solution can be ignored.
Amikacin is an aminoglycoside antibiotic used to treat different types of bacterial infections. Amikacin works by binding to the bacterial 30S ribosomal subunit, causing misreading of mRNA and leaving the bacterium unable to synthesize proteins vital to its growth.
