Tartrate-resistant acid phosphatase 5b (TRACP5b) has been recognized as a marker of osteoclasts. There are two isoforms of type 5 TRACP in human serum, 5a and 5b. Serum TRACP5b is a proteolytically cleaved form with disulfide-linked polypeptide subunits of 16 and 23 kDa. One important difference between these two isoforms is the presence of sialic acid in 5a but not in 5b. Purified human osteoclastic TRACP is type 5b.
