Description:
Guanosine 5'-monophosphate disodium salt (Guanosine 5'-monophosphate disodium salt) is a colorless to white crystalline or crystalline powder, usually containing 7 molecules of crystalline water. Melting point is not obvious, Browning at 240℃, decomposition at 250℃. It is stable to acid, alkali, salt and heat. Strong moisture absorption, soluble in water (25℃, 25%), pH value of 5% aqueous solution is 7.0 ~ 8.5, slightly soluble in ethanol, acetone and ether.
The production of Guanosine 5'-monophosphate disodium salt mainly includes enzymatic hydrolysis and fermentation, industrial significance of two-step fermentation and biosynthesis and chemical synthesis in fermentation.
(1) RNA hydrolysis. See production method for 5 '-inosine disodium.
(2) two-step method. With glucose as carbon source, the strain of bacillus subtilis was used to ferment the guanosine with the production level of 10.5g/L. The guanosine was then acidified in pyridine solution with phosphoric acid trichlorooxyphosphate to obtain guanosine.
(3) biosynthesis and chemical synthesis. At PH=7, bacillus macrosporum (no.336) was used to ferment glucose (8%) for 90h to produce 5-ammonia-4-formamide nucleoside (AICAr) 15g/L. Then it was extracted by ion exchange method, concentrated and dried, dissolved in methanol containing NaOH, and heated with carbon disulfide to change it into 2-thiosinosin. Finally, the Guanosine 5'-monophosphate disodium salt can be obtained by phosphorylation of Guanosine 5'-monophosphate.
