D-glyceraldehyde-3-phosphate ketol-isomerase plays an important role in glycolysis and is essential for efficient energy generation. D-glyceraldehyde-3-phosphate ketol-isomerase has been found in nearly all organisms, including mammals, insects, fungi, plants and most bacteria. But some bacteria that do not undergo glycolysis, such as ureaplasma urealyticum, lack the enzyme.
D-glyceraldehyde-3-phosphate ketol-isomerase was dimer formed by two identical subunits. Each subunit contains about 250 amino acid residues. The three-dimensional structure of each subunit contains eight alpha spirals on the outside and eight parallel beta chains on the inside. Such a pattern, known as an alpha beta bucket or TIM bucket, is the most common form of protein folding ever observed. The active site of the enzyme is located in the center of the barrel, where one glutamate and one histidine participate in the catalytic reaction process. Residues in the vicinity of active sites were conserved in all known d-glyceraldehyde -3-phosphate ketol-isomerase.
