Ampicillin is a beta-lactam antibiotic .Until the introduction of ampicillin by the British company Beecham, penicillin therapies had only been effective against Gram-positive organisms such as staphylococci and streptococci. Ampicillin (originally branded as 'Penbritin') also demonstrated activity against Gram-negative organisms such as H. influenzae, coliforms and Proteus spp. Ampicillin was the first of a number of so-called broad spectrum penicillins subsequently introduced by Beecham. Ampicillin is part of the aminopenicillin family and is roughly equivalent to its successor, amoxicillin in terms of spectrum and level of activity. It can sometimes result in reactions that range in severity from a rash (in the case of patients that may unwittingly have mononucleosis) to potentially lethal allergic reactions such as anaphylaxis.
To produce Ampicillin, the side-chain carboxylic acid of D(-) -phenylglycine was first treated with the chlorination agent PCI5. Make acyl chloride and condense with 6-APA. Add acetone and water in the reaction tank, and add 6-apa when the temperature drops to -5--10℃. Add phenylglycyl chloride hydrochloride, and adjust the pH to 3.5 with 10% sodium hydroxide after the reaction for 0.5h. The reactants were extracted with toluene. Water intake layer, adjust pH to about 3.0 with 10% ammonia. Decolorize with activated charcoal and filter. The filtrate was adjusted with ammonia to a pH of 4.8. It was allowed to sit, then filtered, washed with acetone, and vacuum-dried below 40℃ for Ampicillin.
