3,3',5 '-Tetramethylbenzidine dihydrochloride was used in ELISA reaction as a substrate of catalase, which was capable of producing soluble light blue end products, which could be read by spectrophotometer at 370 or 620-650 nm. 3,3',5 '-Tetramethylbenzidine dihydrochloride reaction can be terminated with 2 M H2SO4 (producing yellow) and read at 450 nm. Therefore, 3,3',5,5'-Tetramethylbenzidine dihydrochloride is a sensitive and specific reagent for the determination of blood, heme and peroxidase.
3,3',5 '-Tetramethylbenzidine dihydrochloride is a new safe reagent. Compared with the conventional chromogenic agents, such as benzidine, o-toluidine and o-phenylene diamine, 3,3',5,5' -tetramethylbenzidine dihydrochloride have the advantages of high detection sensitivity, good stability, and safe use. The results of animal test and Ames bacterial autovariability test indicate that 3,3',5,5'-Tetramethylbenzidine dihydrochloride is a non-carcinogenic and non-mutagenic agent. At present, 3,3',5,5'-Tetramethylbenzidine dihydrochloride has gradually replaced the strong carcinogen benzidine and other carcinogenic benzidine derivatives, and is used in clinical testing, forensic examination, criminal investigation and environmental monitoring.
